Thiophosphoramidites are unique reagents that are used to replace the two non-bridging oxygen atoms with sulfur atoms in a DNA or RNA phosphodiester linkage creating a phosphorodithioate (PS2) linkage. AM sells these reagents through Glen Research Corporation.
• Like natural DNA (RNA), the phosphorodithioate linkage is achiral at phosphorus
• Eliminates stereoisomer formation associated with monothioate (PS) backbones
• PS2 analog is completely resistant to nuclease degradation
• PS2-ODNs bind proteins with a higher affinity than their phosphodiester analogues
• PS2 siRNA enhance gene silencing and increase RNA in vivo stability
